What is required to build them? Does it take current unit supplies or national supplies?

You need 4 elements of supply wagon or transportships. If it is possible use the transportships, they are cheaper.

Just remember that there needs to be a chain to that depot, either by river or by being two to three regions away from the nearest part of a depot chain.
